Histochemical analysis of polarizing colors of collagen using Picrosirius Red staining in oral submucous fibrosis.

چکیده
BACKGROUND Oral Submucous fibrosis (OSMF) is a common potentially malignant disease, which is characterized by generalized fibrosis of the oral soft tissues. It is due to disproportion in the collagen deposition and degradation. The excess deposition of collagen results in juxta epithelial fibro elastic changes and epithelial atrophy. Neoplastic changes in the epithelium usually precede the changes in the connective tissue. The purpose of the study was to evaluate the changes in birefringence of collagen using picrosirius stain and compare with haematoxylin and eosin stained section. MATERIALS & METHODS The study was conducted on 50 subjects, which included 45 patients diagnosed with different functional and histopathological grades of OSMF and 5 in control group. Picrosirius red stain was used to analyze collagen both histopathologically and qualitatively using polarizing microscopy. Chi-square test at p value <0.05 was used to analyze the results and the significance was tabulated. RESULTS Collagen fibers showed mixed birefringence with a shift in polarizing colour from yellow to red-orange in lamina propria, around the muscle and blood vessels, which was correlating with the conventional H&E stain. CONCLUSION The results of present study show a significant change in birefringence of collagen between the various components of connective tissue. This change in birefringence colors and arrangement of collagen fibers might give an assumption of impending neoplastic change in OSMF. متن کاملComparative Study of Clinical Staging of Oral Submucous Fibrosis with Qualitative Analysis of Collagen Fibres Under Polarized Microscopy
Background & Objectives: Oral submucous fibrosis (OSMF) is a condition where excessive deposition of dense collagen fibers occurred in the connective tissue of oral mucosa. An alteration of collagen necessitates an in depth understanding of collagen in oral tissues as no breakthrough studies have been reported. Therefore the aim was to correlate the clinical, functional and histopathol...
متن کاملClinicopathologic study of a series of giant cell fibroma using picrosirius red polarizing microscopy technique.
BACKGROUND Giant cell fibroma (GCF) is a distinct type of fibroma with characteristic large, stellate mononuclear or multinucleated giant fibroblasts; the stroma of GCF is relatively unexplored. The Picrosirus red polarizing microscopy technique is used to characterize the collagen fibers. The aim of this study was to evaluate the staining properties of collagen fibers in GCF and to correlate i...
متن کاملHistochemical Analysis of Collagen Reorganization at the Tumor-Stromal Interface in Oral Squamous Cell Carcinoma- A Polarizing Microscopic Study
Stromal changes are the key factors for provision of nutrition and growth to any tumor and also they act as a barrier for spread of the tumor. There are limited histochemical studies in the literature on methods to detect, quantify and analyze the collagen in the invasive tumor front (ITF) of oral squamous cell carcinoma (OSCC). Aim: 1) To analyze the nature of collagen (thickness, hue, density...
متن کاملSubmucosa Precedes Lamina Propria in Initiating Fibrosis in Oral Submucous Fibrosis - Evidence Based on Collagen Histochemistry
Oral submucous fibrosis is a chronic insidious disease and a well-recognized potentially malignant condition of the oral cavity. It is a collagen related disorder associated with betel quid chewing and characterized by progressive hyalinization of the lamina propria. Objectives: It is traditionally held that in oral submucous fibrosis the hyalinization process starts from the lamina propria and...
